چکیده
Transcription of the psbD-psbC gene cluster in tobacco chloroplasts has been studied. This cluster contains in linear sequence the overlapping genes encoding the D2 and 43 kDa proteins of Photosystem II (psbD and psbC, respectively), and ORF62. Eight major transcripts ranging from 1.5 to 4.4 kb were detected by northern blot analysis. S1 mapping experiments revealed that these multiple transcripts comprise five distinct 5' ends whose precise positions were further determined by primer extension analysis. Two of the five 5' ends were determined to be the transcriptional initiation sites by in vitro capping assays: the main site is located 905 bp upstream from the ATG codon of psbD and the additional site is 194 bp upstream from the first ATG codon of psbC. The latter site and the preceding prokaryotic promoter motif are within the protein-coding region of psbD. The 3' ends of transcripts were determined by S1 mapping.

A Nove1 Light-Regulated Promoter and Dicot Chloroplasts Conserved in Cereal
The chloroplast psbD-psbC genes encode D2 and cp43, a reaction center protein and chlorophyll-binding antenna protein of photosystem II, respectively. We have previously shown that differential accumulation of light-induced psbD-psbC mRNAs in barley chloroplasts is due to transcription from a blue light-responsive promoter (LRP). It is hypothesized that the light-induced mRNAs help to maintain ...
متن کاملldentification of a Sequence-Specific DNA Binding Factor Required for Transcription of the Barley Chloroplast Blue Light-Responsive psjbD-psbC Promoter
The plastid gene psbD encodes the photosystem II reaction center chlorophyll protein D2. psbD is located in a complex operon that includes psbC, psbK, psbl, orf62, and tmG. The operon is transcribed from at least three different promoters. One of the psbD promoters is differentially activated when plants are exposed to blue light. In this study, the psbD blue light-responsive promoter was accur...

The transcription of rice plastid psbD-psbC genes encoding photosystem II reaction center protein D2 and chlorophyll alpha-binding protein CP43 is closely regulated by light. To elucidate the sequence requirement for the light-responsive promoter of psbD-psbC operon, transcriptional analysis of the rice promoter was performed with deleted mutants and site-directed mutants in vitro. Deletion of ...
متن کاملTranslation of partially overlapping psbD-psbC mRNAs in chloroplasts: the role of 5′-processing and translational coupling
The chloroplast psbD and psbC genes encode the D2 and CP43 proteins of the photosystem II complex, and they are generally cotranscribed. We report studies on the basic translation process of tobacco psbD-psbC mRNAs using an in vitro translation system from tobacco chloroplasts. The primary transcript has an unusually long 5'-UTR (905 nt). We show that it is translatable. Processing of the 5'-UT...
متن کاملNucleotide sequence of the barley chloroplast psbC gene.
Nucleotide sequence of the barley chloroplast psbC gene We present here the eequenoe of psbC gene from barley ohloroplaBt genome, ooding for the 43 kDa photoBjrBtem II protein (see alBO CarlBberg ReB.Commun., 49:647 for the last 500 b.p.). The 5'-end of psbC overlaps with the 3'-end of previously sequenoed psbD gene (Nuo Acids Res. 16:5686). The deduced amino acid Bequenoe is presented.
